Experimental Studies on the Changes of Immunoglobulins by Immunoelectrophoresis in Clonorchiasis


Abstract
With the development of immunoelectronhoresis by which more accurate analysis of the serum protein became possible, the changes of immunoglobulins in the various diseased states were investigated.
But none of paper investigated the changes of immunoglobulins and compared the results with the serum 7--globulin Ievels-_in the cases of clonorchiasis sinensis was reported.
The purpose of this study, then, was to investigate the changes of immunoglobulins in experi- .entally produced clonorchiasis and to compare the results with the serum 7--globulin levels.
Thirty six rabbits, 2 kg in weight, were deviled into 7 groups of which 6 rabbits were Infested with 100 metacercariae of clonorchis sinensis (group A), 6 were with 200 (group B), 6 were with 300 (group C), 5 were with 600 (group D), 5 were with 1,000 (group E), and 4 were with 1,500 metacercariae(group F). And 4 animals were treated with carbon tetrachloride by inhalation for 20 minutes every day and served as the CC14-induced damaged-liver group(group G).
The experimental duration and the observed adult worms of clonorchis sinensis were as shop=; n on table 1.
Ianmunoelectrophoretic analyses of the serum collected on a week¢¥s interval were performed by Scheidegger¢¥s modified microimmunoelect-rophoretic method. Each serum antigen sample was run in duplicate. In the first run the serum was in the left hand well and in the second run it was in "kthe right hand in order to avoid inconsistencies in the agar preparation which might affect the position of the precipitating lines.
The antiserum used in this experiment was obtained by immunization for goat against pooled normal rabbit serum.
Microscope slide agar-gel electrophoresis for serum protein fractionation was performed at VC using barbital buffer, pH 8.6 and ionic strength 0.05, with 140 volts and 20 mA for 40 minutes. The fractionated protein was stained with amidoschwar z 10 B and scanned by Spinco Analytrol.
The normal immunoelectrophoregrams exhibited IgA, IgM, and lgG which were strictly same region with those of normal human serum. And the author was able to find out the other protein fraction rr_i.¢¥rated in the immunoelectrophcretic pattern between the IgG and IgA area, and presented the precipitati.--g arc along the IgG on the immunoelectrophoregrams. The author designated this protein fraction of rabb` serum f¢¥71-x" arbitrarily.
The results obtained were summarized as follows:
1. Marked increase of lgM with slight increase of "7i-x" in clonorchiasis were observed, on the other hand marked increase of IgA without the changes of IgNI were observed in the CCI4-induced damaged-liver group in which the "rz-x" was decreased on the late of experiment.
-2: The-increase of IgM and "ri x" in clonorchiasis were closely correlated with the degree of infestation of clonorchis sinensis.
3. There were no decreased cases of IgM in the mild infestation group of clonorchis, o.herwise decreased IgA and IgM were observed on the late of experiment in the heavy infestaticn group.
4. No close correlation between the changes of immunoglobulins and the increase of serum ;r-globulin levels were observed.
